چکیده

More than 5035 of tracer rubidium entering the coronary circulation is actively taken up by the heart muscle cells during a single passage. Rubidium ions must traverse the capillary wall, the interstitial space, and the muscle cell membrane to enter the very large potential rubidium pool within the cells. The aim of this investigation was to determine which, if any, of these steps are rate limiting. The anterior descending branch of the left common coronary artery was perfused at its origin. Pulse injections containing Rb + , Na or C-sucrose, and I-albumin were made into the perfusion line and timed serial samples of coronary sinus blood were collected and analyzed. A model was designed which incorporates rate constants describing both the exchange of Rb at the capillary wall and its entry at the muscle cell membrane. Labeled sucrose or Na + was used to determine flow per interstitial fluid volume, a parameter necessary for the application of the model. It was assumed that the interstitial fluid volume available for labeled sucrose or Na was identical to that available for Rb + . The rate constant describing exchange at the capillary wall (the permeability surface product per unit accessible interstitial fluid volume) increased with perfusion, whereas that for uptake by the myocardial cells was relatively constant and independent of flow.

Evaluation of Cardiac Mitochondrial Function by a Nuclear Imaging Technique using Technetium-99m-MIBI Uptake Kinetics

Mitochondria play an important role in energy production for the cell. The proper function of a myocardial cell largely depends on the functional capacity of the mitochondria.  Therefore it is necessary to establish a novel and reliable method for a non-invasive assessment of mitochondrial function and metabolism in humans. Although originally designed for evaluating myocardial perfusion, 99mTc...

Effect of Branched-Chain Amino Acid Supplementation on O2 Uptake Kinetics and Time to Exhaustion in Trained Women

Objective The aim of this study was to investigate the effect of Branched-Chain Amino Acid (BCAA) supplementation on O2 uptake kinetics and the time to exhaustion in trained women.  Methods In this quasi-experimental study with pre-test/post-test design, participants were 20 trained women (Mean±SD age, 21.3±0.5 years). They were randomly assigned into two groups of BCAA (received 45mg/kg/d BCA...

Age dependence of myocardial Na+-K+-ATPase activity and digitalis intoxication in the dog and guinea pig.

Infants and young animals tolerate higher doses of digitalis glycosides, relative to body weight, than adults. One possible explanation for this could be an age-dependent difference in the myocardial digitalis receptor, the Na+-K+-ATPase. Two functions of this enzyme were studied in adult, 1- and 6-week-old dogs and guinea pigs: in vitro myocardial uptake of rubidium (86Rb) and binding of ouaba...

Myocardial monovalent cation transport during the quinidine-digoxin interaction in dogs.

To study the relationship of the serum digoxin concentration to the digoxin effect on monovalent cation transport during the quinidine-digoxin interaction, we used radiolabeled rubidium to measure monovalent cation active transport in myocardial biopsy samples from dogs. In a preliminary study, we showed that quinidine did not affect rubidium uptake by myocardial samples from intact dogs. Then,...
